Latest research on Cidofovir

Cidofovir is an injectable antiviral medication for the treatment of cytomegalovirus (CMV) retinitis in patients with AIDS. It suppresses CMV replication by selective inhibition of viral DNA synthesis. [Wikipedia]

Topical application with the cytotoxic anti CMV drug Cidofovir has been used to treat CIN2/3 and showed ~60% clearance of CIN but did not eliminate HPV infection as assessed by the hybrid capture 2 assay [31]. [source, 2016]
This prodrug recipe has been employed for various drugs such as the antivirals Valacyclovir (Ganapathy et al., 1998), Valganciclovir (Sugawara et al., 2000), and Cidofovir (McKenna et al., 2005). [source, 2016]
For these experiments, we used toremifiene citrate (TOR; Sigma), Cidofovir hydrate (CDF; Sigma) favipiravir (FPV, T-705; Cellagen Technology), Lamivudine (3TC; Sigma) Zidovudine (AZT), Tenofovir (TFV) maraviroc (MVC; NIH AIDS Reagent Program), Infergen (IFN Alfacon-1, Pharmunion Bsv Development Ltd.) or human interferon beta-1a (IFN-β, Avonex; Biogen). Known substrates for OAT1 include cyclic ADENOSINE monophosphate, cyclic guanosine monophosphate, antiviral agents (Acyclovir, Cidofovir, and Zidovudine), antibiotics, and diuretics [15]. [source, 2016]
Therefore, drugs such as Cidofovir and adefovir may cause concentration-dependent renal toxicity [19]. [source, 2016]
Because Cidofovir, adefovir, and Tenofovir are all nucleotide analogues, Tenofovir accumulation within tubular epithelial cells also may interfere with renal function. [source, 2016]
The most common mutation (UL97) usually appears in isolation without cross-resistance to the other antivirals licensed for CMV treatment, that is, Cidofovir or Foscarnet. [source, 2016]
However, the combination of UL97 and UL54 mutations usually confers cross-resistance to Cidofovir. [source, 2016]
The CMVIG and antiviral agents have complementary modes of action: CMVIG eliminates virus particles before they reach the host cell, whereas antiviral agents (Ganciclovir, Foscarnet and Cidofovir) target viral DNA polymerase to block viral replication within the cell. [source, 2016]
There are no approved treatments, but Ganciclovir, Foscarnet, and Cidofovir have been used. [source, 2016]
